The ‘progressive’ Dems he is targeting (and their growing base) are in favor of pursuing what they see as a more balanced approach to the Israel-Palestinian conflict, and perhaps even a rolling back the alliance with Israel altogether, while the ‘establishment’ Dems are happy continuing the policies of past (pre-Trump) administrations which differed little in their commitment to the alliance. Beyond a minuscule element of the Libertarian side of the party, the Republicans are absolutely united in their commitment to Israel. So it’s an easy way for Trump to stoke division within the rival party.
